Seperti pada seri Generasi Advanced dan Diamond & Pearl sebelumnya, pada serial Pokemon Best Wishes! seri dimulai dengan Ash Ketchum, dia berangkat ke wilayah Unova yang terletak jauh dari Kanto, Johto, Hoenn dan Sinnoh, dengan Pikachu nya.
